The Penken Trophy begins with a flat warm-up section through the centre of Mayrhofen, heading west, then downstream along the Ziller River and on a gravel forest path to the start of the first ascent. On a steep path we first go steep, then a bit flatter through a forest. After a flatter section, the second ascent begins over steep trails. The path then leads into a flatter asphalt road. Past the Gschößwandhaus and lush meadows, the route leads to the first destination at the Penkenbahn mountain station. The final two kilometres run on a gravel road slightly uphill to the end point, the Penkenalm.
